Proliferation Assay

Proliferation response is a powerful functional to determine pathogen exposure or graph rejection. In the past, proliferation has been accomplished by 3H- Thymadine or BrdU incorporation. Over the years, researchers have used Carboxyfluorescein diacetate succinimidyl ester (CFSE) incorporation for easy cell labeling and analysis. CFSE passively diffuses into cells reacts with intracellular amines, forming fluorescent conjugates retained within the cell. When cells proliferate, fluorescent intensity is diminished allowing researchers to accurately quantitate cellular response.

 

IQFCS recently optimized proliferation assay to phenotype specific lymphocyte subsets response to antigen stimulation. IQFCS optimized antibodies conjugated to non-bleaching fluorescent tags providing a clear  separation between positive and negative cells.

Human PBMCs were purified, CFSE stained, and stimulated with ConA and incubated for 48 hours. Thirty minutes prior to analyzing, cells were stained with human anti-CD8-AQ-647 and analyzed for proliferation. Using the optimized proliferation kit allowed for easy quantification of proliferative response by CD8-low and CD8-high lymphocytes.  

IQFCS offers three different proliferation kits: "Basic" kit contains CFSE only, "Basic Plus" kit contains CFSE and hIgG-AQ647 secondary antibody allowing researchers to use other primary antibodies. "Total Proliferation" kit contains CFSE and AQ647 conjugated to hCD3, 4, 8, or mu.
