• A programmed process known as APOPTOSIS or
• Direct lysis of the cell membrane known as NECROSIS.
In this example, the mechanisms of cell death initiated by two different proteins were examined. Either protein A or B was incubated with an epithelial cell line for 24 hours. Thirty minutes before analysis, a cell permeant apoptosis probe was added. Cells were then released from the plate; a live-dead stain was introduced and analyzed by flow cytometry. A necrotic response was induced by protein A while protein B initiated apoptosis. This assay allows the scientist to easily determine which mechanism a compound uses to cause cellular death.
